About Stoke:

Stoke Therapeutics (Nasdaq: STOK), is a biotechnology company dedicated to addressing the underlying cause of severe diseases by upregulating protein expression with RNA-based medicines. Using the company’s proprietary TANGO (Targeted Augmentation of Nuclear Gene Output) approach, Stoke is developing antisense oligonucleotides (ASOs) to selectively restore protein levels. Stoke’s first compound, STK-001 is in clinical testing for the treatment of Dravet syndrome, a severe and progressive genetic epilepsy. Dravet syndrome is one of many diseases caused by a haploinsufficiency, in which a loss of ~50% of normal protein levels leads to disease. Stoke is pursuing the development of STK-002 for the treatment of autosomal dominant optic atrophy (ADOA), the most common inherited optic nerve disorder. Stoke’s initial focus is haploinsufficiencies and diseases of the central nervous system and the eye, although proof of concept has been demonstrated in other organs, tissues, and systems, supporting the company’s belief in the broad potential for its proprietary approach. Stoke is headquartered in Bedford, Massachusetts with offices in Cambridge, Massachusetts.

Position Purpose:

The Bioanalytical, DMPK and Biomarker Development team is seeking an enthusiastic, collaborative and experience Bioanalytical Scientist to join their team. This position will report to our Associate Director of DMPK and is located in our lab facility at 45 and 47 Wiggins Avenue, Bedford, MA.

Key Responsibilities:


  • Developing high-quality assays to quantitate oligonucleotides in biological matrices to understand the pharmacokinetics (PK) and metabolism of drug candidates.
  • Responsible for sample analysis to support PK/PD and other non-clinical studies, interpret results and report data to support pipeline.
  • Partner with and support Pharmacology and Discovery Biology teams.
  • Conduct oligonucleotide stability profiling and metabolite identification studies.
  • Work with CRO laboratories to transfer, qualify/validate bioanalytical assays, and support assay implementation.
  • Write technical reports as needed.
  • Present results at cross-functional team and lab meetings.
  • Maintain accurate documentation of experiments in ELN.
  • Help maintain the bioanalytical lab and keep the instruments running properly.


Required Skills & Experience:

  • At Scientist level: Bachelor’s degree with 10+ years of experience, or Master’s degree with 8+ years of post-graduate experience, or Ph.D. with a minimum of 2 years of relevant experience working in industry or academia.
  • Proven experience with LC-MS platforms such as QqQ and HRMS (Sciex/Thermo) for quantitation of drug molecules.
  • Experience using immunoassay techniques like ELISA, MSD, Western blotting etc. will be a plus.
  • Strong knowledge of bioanalytical principles is required. Experience in metabolism aspect of oligonucleotides is desirable.
  • Good understanding of bioanalytical assay development, validation, and implementation.
  • Excellent problem-solving skills and ability to work as a team player in a dynamic environment.
  • Excellent communication and presentation skills required.
  • Proficient in standard Microsoft Office tools: Word, Excel, GraphPad and PowerPoint.
  • Strong attention to detail and organizations skills.
  • Prior drug development experience in nucleic acids-based therapeutics will be plus.
  • Basic understanding of GLP with ICH and FDA Guidance is a plus.

Travel:

This position will require approximately 0-5% travel.
